Normalmente la mitocondria procesa la glucosa produciendo piruvato y di\u00f3xido de carbono v\u00eda la glicolisis, por el proceso de fosforilaci\u00f3n oxidativa.\u00a0<\/p>\n<p>&nbsp;<\/p>\n<p>En la c\u00e9lula cancerosa el metabolismo de la glucosa se reprograma y la producci\u00f3n de energ\u00eda se desarrolla principalmente por glicolisis aer\u00f3bica que ocurre principalmente debido a la sobreexpresi\u00f3n del gen GLUT1, cuya funci\u00f3n permite importar una mayor cantidad de glucosa al citoplasma, lo cual se ha convertido en un \u00fatil indicador de actividad tumoral en estudios de PET. Se sabe que la activaci\u00f3n de oncogenes, especialmente RAS y MYC, as\u00ed como mutaciones en genes supresores tumorales como TP53, le permite a la c\u00e9lula cancerosa incrementar su proliferaci\u00f3n, evadir los controles citost\u00e1ticos e inhibir los mecanismos de apoptosis. Las c\u00e9lulas tumorales se ven expuestas a condiciones de hipoxia, lo cual, a su vez, aumenta la expresi\u00f3n de enzimas glucol\u00edticas y transportadores de glucosa, incrementando la gluc\u00f3lisis.<\/p>\n<p>Entre los complejos sistemas celulares de respuesta, se encuentran los factores de transcripci\u00f3n HIF1 alfa y HIF2 alfa, que responden a la hipoxia estimulando la gluc\u00f3lisis. Se agrega a este est\u00edmulo la oncoprote\u00edna RAS que interviene independientemente. Sin embargo; dado que la producci\u00f3n de ATP es menor en condiciones de hipoxia, la c\u00e9lula cancerosa requiere que se active la fosforilaci\u00f3n oxidativa mitocondrial, para lo cual es necesario un mayor aporte de ox\u00edgeno, lo cual se compensa con la angiog\u00e9nesis en desarrollo.<\/p>\n<p>Por otra parte, la gluc\u00f3lisis activa mecanismos alternativos hacia otras v\u00edas que sintetizan nucle\u00f3sidos y amino\u00e1cidos, favoreciendo la bios\u00edntesis de macromol\u00e9culas que aportan sustrato para el crecimiento tumoral.
