La mayor\u00eda de las veces no es de car\u00e1cter hereditario, sino que se desarrolla in situ, como consecuencias de la alta tasa de mutaciones que ocurre en el tejido mamario. Principalmente las mutaciones consisten en acumulaciones progresivas de diferentes tipos, tales como mutaciones puntuales, amplificaciones cromos\u00f3micas, deleciones, rearreglos, translocaciones y duplicaciones.<\/p>\n<p>&nbsp;<\/p>\n<p>Se han identificado algunos factores epigen\u00e9ticos que da\u00f1an la estructura del ADN produciendo dichas mutaciones, entre ellas est\u00e1n el humo del tabaco, los rayos UV, productos alquilantes, varios componentes qu\u00edmicos e incluso desechos del propio metabolismo celular. Todos ellos son capaces de producir da\u00f1o al ADN en la modalidad de rupturas de cadena sencilla y de doble cadena del ADN.<\/p>\n<p>Como es sabido, los genes BRCA1, BRCA2 son genes supresores tumorales, que codifican para prote\u00ednas de reparaci\u00f3n de da\u00f1o al ADN, participan en la regulaci\u00f3n de otros genes e intervienen en los procesos de recombinaci\u00f3n entre secuencias hom\u00f3logas del ADN.<\/p>\n<p>Un paso esencial en el proceso de reparaci\u00f3n por recombinaci\u00f3n hom\u00f3loga es la conformaci\u00f3n del llamado complejo MRN, el cual est\u00e1 formado por una mol\u00e9cula de ATM, por el gen Rad50 y por prote\u00ednas que, en conjunto con endonucleasas, consolidan la uni\u00f3n con la prote\u00edna Rad51. El gen ATM, considerado como de moderada susceptibilidad para el c\u00e1ncer de mama, tiene un gran potencial oncog\u00e9nico, ya que se trata de un gen con capacidad de detectar rupturas de doble cadena y activar numerosas prote\u00ednas involucradas en el control del ciclo celular y reparaci\u00f3n del ADN, por lo que sus alteraciones estructurales dejan desprotegida a la c\u00e9lula contra eventuales mutaciones.<\/p>\n<p>En el c\u00e1ncer de mama tambi\u00e9n est\u00e1n involucrados muchos otros mecanismos moleculares, como los que participan en la v\u00eda FANC\/BRCA, en la que, junto con 11 prote\u00ednas, se activa el sistema reparaci\u00f3n por recombinaci\u00f3n hom\u00f3loga, respondiendo a la presencia de variantes patog\u00e9nicas.<br \/>La heterogeneidad observada en el c\u00e1ncer de mama es amplia y cada vez m\u00e1s se descubren mecanismos moleculares en los que participan m\u00faltiples genes.<\/p>\n<\/figure>\n<\/div>\n<\/div><\/div>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<\/div>","protected":false},"excerpt":{"rendered":"<p>Se sabe que el c\u00e1ncer de mama ocupa una alta frecuencia en todas las poblaciones del mundo.
